Screening of various plants viz; Oreganum vulgare (Oregano), Ocimum basilicum (Basil), Cymbopogon citratus (Lemongrass), Corriander sativum (Corriander), Mentha piperita (Mint), Trigonella foenumgraceum (Fenugreek) and Aloe barbadensia (Aloe Vera) have been screened for the presence of protease enzyme.
4.1 Screening of various plants for protease enzyme Protease has been extracted from various plant samples and assayed for enzyme activity using standard protocols as mentioned in materials and methods. Table 1 shows the specific activity of the various plant sources studied.
4.2 Partial Purification of protease isolated from O. basilicum O. basilicum (Basil) extract was partially purified by various downstreaming techniques mentioned below.

4.3.2 pH optima The pH of the reaction mixture containing protease was varied from 3-10. The enzyme was found to show maximum activity at pH 5 (Figure 4.3) in contrary with the results obtained by Vidyalakshmi and Selvi.

4.3.3 Temperature Optima The optimum temperature for activity of protease enzyme isolated from O. basilicum was determined by varying the temperature from 10-900C. In contrary to Lakshmi and Prasad the protease enzyme isolated from O. basilicum was found to show maximum activity at 300C.
