Title: Wet mount
Aim: The aim of this experiment is to make a wet mount side and observe the paramecium’s and other microorganisms found in the pond water.
Background information/ introduction
A technique that is widely used in science biology is wet mounts. Practising this technique helps in order to study slides with specimen under a microscope. In this skill the specimen ejected along with a drop of liquid (e.g. water) and then it is covered with a cover slip. Unlike permanent mount slides, wet slides cannot be stocked for over a long period of time as the liquid might evaporate and growth of unwanted microorganisms may lead to the contamination of the slide.

Dye can be used on wet mounts but it’s completely optional. Some particular dyes can be used to discolour the DNA within the cell and lipids.
Materials required Microscopic slides Coverslips Micropipettes Paramecium and pond water cultures
Methods
A clean and scratch free glass slide was taken and the slide was labelled with the name of the organism or the sample. In the middle of the slide 15-20 μl of culture was placed. Diagonally a clean coverslip was lowered and dropped from one side avoiding any air bubbles. The prepared slide was examined under the microscope firstly under 10× the following by 40× and 100× magnification. Observations were drawn and any mobility was recorded. The cells and their borders in the wet mounts were not as distinct in appearance as in the staining methods. While the bacteria shapes were able to be identified in the yeast wet mount, it was very difficult to determine the shapes in the cheek smear. The next method used was the direct staining using crystal violet. The cells in the direct stained slides were very easy to view, well defined and were a distinct violet color. The indirect staining method using the Congo Red dye appeared to actually stain the cells in the cheek smear rather than the background and area around the cell. In the higher resolution cheek cell slide, the cell was outlined with the Congo Red dye. We have become so used to this diverse range of bread, mouth watering pastries and cakes that seldom we ask just how they come into existence.
