Bio 203 Lab

Measurement of Acid Phosphatase Activity Under a Variety of Environmental Conditions

Introduction:

Biological enzymes are very important as they allow the human body to function and produce a wide variety of biochemical reactions that help sustain life. The main purpose of this lab is to understand the narrow optimal range and varying perimeters that allow these enzymes to optimally function. The enzyme, acid phosphatase, and the substrate p-nitrophenylphosphate were subjected to several different conditions, such as reaction time, varying amount of substrate concentrations, differing amounts of pH and varying reaction temperatures. The overall prediction for this lab would be that enzymes have a very limited range which they optimally function at, and by measuring the absorbance through a spectrophotometer the farther away from biological (human body conditions) the less the absorbance/ products would be in solution.

Materials and Methods:

All of the portions of this experiment have a full list of materials and methods provided in the lab …show more content…
6.0mL - 1.5mL = 4.5 mL

Figure 2: Reaction as a function of time vs absorbance.

Figure 2: The reaction was maintained at 37*C by heat blocks, it seems as though heat may have damaged the 60 minute reaction as the heated was unable to maintain a steady temperature and fluctuated quite a bit so the enzyme may have been damaged in the last reaction.

example calculations: y = mx+b y= .8753x + .0435
.018 = .8753x + .0435
-.0255 = .8753x x=-.0291 Figure 3: [p-nitrophenol] formed, mM vs time

Applying these uses of chicken feathers to concrete could increase the strength and life of the structure built; for example, an office building could potentially last years longer if the concrete used to construct it (contained more strength) to hold up the structure. Furthermore, the wear and tear of the strength of the building would be approximately the same; however, the capacity to support the structure would be greater due to an increased initial strength (Fan, 2008). Worrell, Price, Martin, Hendricks, and Meida (2001) reported that approximately five percent of the global carbon dioxide emissions come from the pouring of Portland cement. Consequently...

Study in Pechay

...Introduction Banana (Musaspp.) is the fourth most important global food commodity after rice, wheat and maize in terms of gross value production. At present, it is grown in more than 120 countries throughout tropical and subtropical regions and it is the staple food for more than400 million people (Molina and Valmayor, 1999). Among the production constraints, Fusarium wilt caused by the fungus Fusarium oxysporum f.sp cubense(Foc) is the most devastating disease affecting commercial and subsistence of banana production through out the banana producing areas of the world (Ploetz, 2005). The disease is ranked as one of the top 6 important plant diseases in the world (Ploetz & Pegg, 1997). In terms of crop destruction, it ranks with the few most devastating diseases such as wheat rust and potato blight (Carefoot andsprott, 1969). The disease almost destroyed the banana export industry, built on the Gros Michel variety, in Central America during the 1950’s (Stover, 1962). In addition, the widely grown clones in the ABB ‘Bluggoe’ and AAA ‘Gros Michel and Cavendish’ sub groups are also highly susceptible to this disease worldwide. Presently, Fusarium wilt has been reported in all banana growing regions of the world (Asia, Africa, Australia and the tropical Americas) except some islands in the South Pacific, the Mediterranean, Melanesia,and Somalia (Stover, 1962; Anonymous, 1977; Ploetz and Pegg, 2000). The fungus Focis the soilborne hyphomycete and is one of more than 100 formae...
