Plating the cells in and after incubation, next day we treat the cells with the compounds; concentration 50uM, and we keep them 24h. Next day washing with PBS and fixation with glutaraldehyde (2.5% in 0.1 M cacodylate buffer or phosphate-buffered saline, overnight). Lavage and wash the cells by twice, then we started to scratch the cells and then centrifugation at 1000g for 10 minutes. Fix the cell pellets with a freshly prepared solution 1.3 % (w/v) osmium tetroxide in colliding buffer for 1-2 hour, then we dehydrate the fixed cells by successive passage through 25, 50, 75 and 95 % solutions of acetone in water (15-30 minutes each).
