Till date Thalidomide’s exact mode of action, essential to its efficacy is unclear and research in the last decade has led to the proposal of 16 possible mechanisms. Details of Thalidomide’s molecular mechanism and its molecular targets can help to reveal analogues lacking the teratogenic effects. This report will particularly focus on some of the recent studies into Thalidomide’s teratogenic mode of action on malignant MM cells; specifically its mode of action on cereblon (CRBN) in MM. Results Studies have revealed that Thalidomide directly binds with CRBN, a protein that forms a complex with E-3 ubiquitin ligase activity along with its binding partners, cullin-4A (CUL4A), DNA-binding protein-1(DBP1) and regulator of cullins-1(ROC1). When Thalidomide binds to CRBN, it is thought that CRBN recruits and binds substrate proteins in this complex resulting in the ubiquitin-proteasome pathway of the proteins, Ikaros (IKZF1) and Aiolos (IKZF3). These transcription factors are essential for B cell, T cell and plasma cell development. Supporting this theory, Thalidomide-resistant MM patient cells were analysed retrospectively. Results showed that patients had suppressed levels of CRBN, and thus suppressed levels of IKF1 and IKF3 in myeloma cells. With this ongoing dilemma directly affecting the students and faculty, I propose that USC opens another parking garage on campus to better accommodate these people. If the University of South Carolina were to build at least two new parking garages on campus, I guarantee that the complaints about the lack of parking on campus would decrease, and attendance by students as well as professors would not be directly affected by the strain of parking spots on campus. As a result of construction of the Patterson dormitory, USC housing has the stressful task of trying to find proper housing for the incoming freshman of the fall semester.
